I.c.v. Infusion of MTII Reduced Food Intake and Body Weight but Increased Oxygen Consumption in Leptin-resistant MSG-obese Rats

Abstract
To evaluate whether MTII, a melanocortin receptor 3/4 agonist, is working in hypophagic, low energy expenditure, and leptin resistant rat model, we measured food intake, body weight, energy expenditure, and fat mass after intracerebroventricular (i.c.v.) infusion of MTII in monosodium glutamate (MSG) induced obese rats. MTII or aCSF was infused into i.c.v. with osmotic minipump for 1 week. MSG-obese rats were induced by neonatal injection (2, 4, 6, 8, 10 th day, 4 mg/kg, s.c.) of MSG. Five-month old MSG rats were characterized by hypophagia, low oxygen consumption rate, reduced leptin sensitivity, and obesity compared with age-matched control rats. Infusion of MTII suppressed food intake and body weight in MSG-obese rats as well as control rats compared with aCSF infused MSG-obese and control rats, respectively. The visceral fat mass was decreased by MT-II infusion in MSG-obese and control rats compared with aCSF infused rats. The oxygen consumption was increased in MSG-obese and control rats compared with pair-fed rats. Surprisingly, the anorexic effect of MTII was greater in MSG-obese rats than in control rats.
Our results suggested that activation of melanocortin system may be used as a treatment modality in already hypophagic, low thermogenic, leptin resistant obese model.
